Hi all,
It works well when it works:) The NEB site (and available software tools like Geneious) have good modules to help design the overhangs.
Need a blunt cutting site on plasmid is all, and about 50bp overhangs on insert.
Key for the process is that you’re going to want to spend money for (or create) really really good competent cells bc it’s a low frequency thing.
