Developing LPS-targeting bacteriocins as strain specific antibacterials
Abstract: Since interactions between microbes can determine assembly dynamics within microbiomes, outcomes of these interactions have the potential to directly shape functional outputs of microbial communities. A deeper understanding of the molecules that drive these interactions may therefore enable fine scale manipulation of microbiomes in both clinical and agricultural settings. Over the last decade, the Baltrus lab has discovered and characterized a variety of molecules produced by bacterial phytopathogens, including tailocins (which are antimicrobials derived from phage tails), which are highly specific and can be used to target and kill numerous agriculturally relevant pathogens. We have further developed multiple assays to characterize the genetic and genomic basis of tailocin production and sensitivity, and we have demonstrated that the lipopolysaccharide layer is the likely the binding site for tailocins. Lastly, we demonstrate the potential for these molecules to be developed into prophylactic antibiotics to protect crops from colonization by clinical and agricultural pathogens.
